Phosphatase Inhibitor Cocktail is a broad-spectrum cocktail designed to protect phosphoproteins from dephosphorylation during lysis and extraction procedures from mammalian cells or tissues and for immunoprecipitation and kinase assays.

The cocktail is provided as a ready-to-use broad-spectrum solution of inhibitors targeting both serine/threonine phosphatases and protein tyrosine phosphatases (PTPs). The product is supplied as a 100X concentrated stock solution in a liquid format for improved accuracy, solubility, and ease of use in comparison to traditional tablets

's Phosphatase Inhibitor Cocktail is a broad-spectrum cocktail designed to protect phosphoproteins from dephosphorylation during lysis and extraction procedures from mammalian cells or tissues and for immunoprecipitation and kinase assays. The cocktail solution inhibits both serine/threonine phosphatases and protein tyrosine phosphatases (PTPs). Phosphatase Inhibitor Cocktail is provided as a ready-to-use 100X stock solution. The cocktail composition and inhibitor targets are listed below:

- Sodium Fluoride - Acid Phosphatases - Sodium Orthovanadate - Protein Tyrosine Phosphatase, Alkaline Phosphatase - Sodium Pyrophosphate - Serine/Threonine Phosphatases - β-Glycerophosphate - Serine/Threonine Phosphatases

It is recommended to use 10 μl of the cocktail solution for every 1 ml of cell lysate. Since phosphatase levels may vary among cell and tissue types, it may be necessary to increase the concentration of inhibitors.

Note: Compounds precipitate out of solution at 4°C. To resuspend, bring the cocktail up to room temperature and shake the solution until any visible crystals completely dissolve into the liquid.

For quality control, each batch of Phosphatase Inhibitor Cocktail is tested for its ability to inhibit alkaline phosphatase activity. (Figure 1) Alkaline phosphatase activity was measured at 0-50 ng/well in the presence or absence of Phosphatase Inhibitor Cocktail. In the presence of Phosphatase Inhibitor Cocktail, no alkaline phosphatase activity is observed, while in the absence of phosphatase inhibitors, increased alkaline phosphatase activity levels are observed with increasing alkaline phosphatase concentrations.
